What is an Estradiol (E2) test? The estradiol test checks the level of the hormone estradiol in the bloodstream. This estradiol plays an important role in sexual development and is the most important form of the hormone estrogen. In a sexually mature female, it is produced mainly in the ovaries and in smaller amounts by the adrenal glands.
